Working Principle Of Fluorometer

FLUOROMETER, or fluorkneter, an instrument used in chemical analysis to measure the intensity of the fluorescence emitted by a substance. It is an extremely Sensitive instrument, detecting quantities of uranium and other fluorescent materials as minute as \displaystyle {{10}^{-9}} gram.

The design of the instrument is based on the fact that a substance emits fluorescence after it absorbs radiant energy from an ultraviolet light source. The fluorometer consists of an ultraviolet light source, a filter that transmits ultraviolet light between the source and the sample to be analyzed, a filter that transmits the fluorescence emitted by the sample, a phototube that detects the intensity of the fluorescence, and a microammeter that measures the current from the phototube. When diffraction gratings or prisms are used instead of filters, the instrument is called a spectrofluorometer.

The ultraviolet light source commonly is a high- or low-pressure mercury vapor lamp or a xenon arc lamp. As shown, the fluorescence usually is measured at a right angle to the ultraviolet light path to avoid interference from the source.
